sequence not suucceful when including std

Dear Sir
Im trying also 3 point of standards of 5 levels and run a sample, but sequence doesnt work and software ask that i have to create a calibration file in the method setup, to link a new calibration file before creating table of peak and concentration? because when a create a new calibration file, in the calibration window, to add all peak is dimmed and not actif and I can see only one pane even setting the windows of calibration to "show both",
please let me know when including in sequence standards, what should I do exactly before concerning a calibration file

Dear Samira,
the warning in Sequence (and inability to run it) is caused by usage of samples marked as calibration standards and no calibration linked to the particular method. Calibration standards measured in the sequence are used to update the calibration file automatically, but when you have none specified, Clarity would not know which one to update.

The solution is to create the calibration file first and link it to the created method. This can be most simply done by going into the Method Setup dialog, Calculation tab of the created method, and pressing the New... button. Clarity will ask to name the new calibration, do so according to your needs. Then, please press the View... button to open the Calibration window (close the Method Setup dialog as you would not be able to modify the Calibration while it is open). There you are supposed to fill the calibration table (manually, either by entering the compounds and their retention times directly, or using some already measured calibration standard, which is the preferred way). The columns you should concentrate on are Compound Name, Retention Time and Amount as bare minimum - other columns will be filled too by default values.

The Amount column must be filled in for each compound in the Calibration Table and for each calibration level - as the amounts of different compounds may differ, it is not possible to fill the Amount field automatically from the Sequence window in Clarity, just the responses are filled.

Generally you are supposed to measure at least one sample before starting any large-scale automatic sequence runs. On this one sample, you should fine-tune the integration parameters (and transfer them back to the Instrument method for newer samples) and create the calibration file (and, once again, link it with the Instrument method). All the basic procedures are described as step-by-step in our online Clarity Guide.

As for the ??? in the Data Acquisition window, this means no analysis is running (thus no analysis time is measured). Once you start the analysis, the time will begin to increment correctly.
